The fire started around 4 a.m. Saturday on the unit bock of West Washington Street.
Mayor Gary Stevenson said police arrived to the scene initially and were told there were children in the burning home.
Officers tried to gain entry into the house, but were driven back by the flames and smoke.
Firefighters responded within five minutes, the mayor said.
Crews put up ladders and were able to rescue two children from the second floor.
The children were unconscious and were taken to the hospital in critical condition where they were revived.
Family members said the woman, her grandbaby and adult son managed to escape before firefighters arrived.
Family members also said the grandmother was babysitting the two injured children but they are not related to her.
The fire was placed under control just before 7 a.m.
The mayor said the first floor of the home was completely gutted.
The Red Cross is assisting the family.
The fire marshal and prosecutor's office are investigating a cause for the fire.
